Human Alpha-1B-glycoprotein/A1BG ELISA Kit

Manufactured by Assay Genie, this Sandwich ELISA Kit is intended for the quantitative measurement of human Alpha-1B-glycoprotein (A1BG). The assay uses a double-antibody sandwich format with colourimetric readout at OD450 and a reaction duration of 120 minutes. It is validated for use with human serum and plasma samples.

According to the manufacturer's specifications, the kit offers a sensitivity of 0.375 ng/mL and a measuring range of 0.625-40 ng/mL. Reported recovery values are 88-104% (average 95%) for serum (n=5), 93-105% (average 98%) for EDTA plasma (n=5) and 87-99% (average 95%) for heparin plasma (n=5). Linearity has been demonstrated by diluting spiked samples at 1:2, 1:4 and 1:8, with serum ranging from 87-105%, 94-104% and 89-104% respectively; EDTA plasma from 86-100%, 82-100% and 82-98%; and heparin plasma from 83-99%, 91-100% and 83-91%. Intra-assay CV values range from 3.87% to 4.35% and inter-assay CV values from 3.51% to 4.69% across low, medium and high concentration samples. Reagents should be stored at 2-8°C.

The procedure involves the sequential addition of capture/detection antibody working solution together with standards or samples, incubation at 37°C, washing, addition of HRP-Streptavidin, further incubation, a second wash, TMB substrate development and a stop step, with absorbance read at 450 nm. Serum is prepared by allowing whole blood to clot at room temperature or 2-8°C followed by centrifugation at 1000 × g for 20 minutes; plasma is collected using anticoagulant tubes such as EDTA-Na2/K2 and centrifuged at 1000 × g for 15 minutes at 2-8°C within 30 minutes of collection.
