Description

From Assay Genie, this Sandwich ELISA kit enables the in vitro quantitative determination of human SERPINA3 (alpha-1-antichymotrypsin) concentrations in serum and plasma samples. It uses a double-antibody, colorimetric sandwich ELISA format and is intended for research use only. The standard curve covers 1.56–100 nmol/ml with a reported sensitivity of 0.938 nmol/ml. Performance characteristics supplied by the manufacturer include intra-assay CV <8% and inter-assay CV <10%.

Alpha-1-antichymotrypsin (SERPINA3, UniProt P01011) is a secreted serine-type endopeptidase inhibitor of the serpin family, mapped to chromosomal region 14q32.1. Although its precise physiological role is unclear, it is known to inhibit neutrophil cathepsin G and mast cell chymase, both capable of converting angiotensin-1 to active angiotensin-2. According to the NCBI summary, SERPINA3 is a plasma protease inhibitor whose polymorphisms appear tissue-specific and influence protease targeting, and variations in its sequence have been implicated in Alzheimer's disease, with deficiency associated with liver disease and mutations identified in patients with Parkinson disease and chronic obstructive pulmonary disease.

The kit is supplied with a dismountable 8×12-strip ELISA microplate, lyophilized standard, sample/standard dilution buffer, biotin-labeled antibody, antibody dilution buffer, HRP-streptavidin conjugate (SABC), SABC dilution buffer, TMB substrate, stop solution, 25× wash buffer and plate sealers, with most components stored at 4°C and the lyophilized standard held at 4°C or –20°C. Recovery ranges from spiked matrix samples are reported as 89–101% for serum, 87–103% for EDTA plasma and 94–103% for UFH plasma, with corresponding averages of 93%, 97% and 98%.
