Human ALDH4A1/Delta-1-pyrroline-5-carboxylate dehydrogenase ELISA Kit

This colorimetric Sandwich ELISA, Double Antibody kit from Assay Genie enables quantitative detection of human ALDH4A1 (Delta-1-pyrroline-5-carboxylate dehydrogenase, mitochondrial; UniProt P30038) in serum, plasma, cell culture supernatant, cell or tissue lysate, and other liquid samples, and is intended for metabolism research.

ALDH4A1 catalyses the irreversible conversion of delta-1-pyrroline-5-carboxylate (P5C), derived from proline or ornithine, to glutamate, providing a step that interconnects the urea and tricarboxylic acid cycles. Dysregulation of ALDH4A1 has been linked to various metabolic disorders and neurological conditions, making it a useful biomarker for research into these areas.

Validated linearity across 1:2, 1:4 and 1:8 dilutions covers serum (87-100%, 86-102%, 85-101%), EDTA plasma (82-101%, 86-101%, 84-99%) and heparin plasma (82-94%, 81-99%, 81-88%). Spike-and-recovery is 97-104% (average 99%) in serum, 91-105% (average 98%) in EDTA plasma, and 91-102% (average 97%) in heparin plasma. The protocol follows a standard sandwich workflow: 90-minute sample incubation at 37°C, 60-minute biotinylated detection antibody, 30-minute HRP-streptavidin (SABC), 10-20 minute TMB development in the dark, and absorbance reading at 450 nm after stop solution. Reagents are stored at 2-8°C for 12 months.
