Description
From Assay Genie, this Sandwich ELISA Kit uses a double-antibody sandwich ELISA format with colorimetric detection and is designed for the in vitro quantitative determination of Human ALAD (Aminolevulinate Dehydratase) concentrations in human serum, plasma, cell culture supernatants, tissue homogenates, and other biological fluids. The kit has a reported sensitivity of 9.375 pg/ml with a working range of 15.625–1000 pg/ml, intra-assay CV < 8%, and inter-assay CV < 10%.
Included components are a dismountable 8×12-strip microplate, a lyophilized standard, sample/standard dilution buffer (20 ml), biotin-labeled antibody (120 µl), antibody dilution buffer (10 ml), HRP-Streptavidin conjugate / SABC (120 µl), SABC dilution buffer (10 ml), TMB substrate (10 ml), stop solution (10 ml), 25X wash buffer (30 ml), and plate sealers (5). Kit components are stored at 4°C (with the lyophilized standard also acceptable at -20°C), and light-sensitive reagents should be protected from light; the kit is stable for 6 months at 4°C.
Spike-and-recovery in serum (n=5), EDTA plasma (n=5), and UFH plasma (n=5) yielded average recoveries of 98%, 96%, and 96%, respectively. Linearity was demonstrated across 1:2, 1:4, and 1:8 dilutions. The kit detects ALAD (UniProt P13716; NCBI Gene ID 210; KEGG K01698), an enzyme involved in heme biosynthesis whose dysregulation has been linked to lead poisoning, porphyrias, and certain cancers.
