Description

From Assay Genie, this Sandwich ELISA kit is designed for the quantitative detection of human AIF1 (Allograft Inflammatory Factor 1) using a double-antibody immunoassay format. The kit is reactive with human samples and validated for use with serum, plasma, cell culture supernatant, cell or tissue lysate, and other liquid sample types. AIF1 is a protein involved in inflammatory responses, immune regulation and tissue repair processes and is studied in relation to autoimmune disease, chronic inflammation and transplant rejection.

The assay has a measuring range of 31.25–2000 pg/mL with a stated sensitivity of 18.75 pg/mL. The protocol follows a sandwich enzyme immunoassay workflow: equilibrate reagents and TMB substrate, incubate standards and samples on the pre-coated plate at 37°C for 90 minutes, add biotin-labelled detection antibody and incubate at 37°C for 60 minutes, add HRP-Streptavidin (SABC) and incubate at 37°C for 30 minutes, develop with TMB substrate in the dark for 10–20 minutes, then add stop solution and read absorbance at 450 nm. Unopened kit components are stored at 2–8°C for 12 months.

Reported linearity values at 1:2, 1:4 and 1:8 dilutions are 85–101%, 87–102% and 86–105% for serum (n=5); 82–101%, 82–96% and 83–93% for EDTA plasma (n=5); and 80–95%, 85–100% and 80–100% for heparin plasma (n=5). Recovery averages are 94% for serum, 97% for EDTA plasma and 94% for heparin plasma. The kit is offered by Assay Genie for research use in cell biology.
