Description

Assay Genie's sandwich CLIA kit enables in vitro quantitative determination of human aryl hydrocarbon receptor (AhR) in serum, plasma and other biological fluids using a chemiluminescence-based detection method. The assay is performed on a microplate pre-coated with an antibody specific to human AhR; standards or samples are combined with the capture antibody, followed by sequential incubation with a biotinylated detection antibody and an Avidin-Horseradish Peroxidase (HRP) conjugate. After washing, a substrate solution is added and the relative light unit (RLU) value is measured on a chemiluminescence immunoassay analyzer, with signal positively correlated to AhR concentration.

The kit recognizes human AhR in samples, with no significant cross-reactivity or interference between human AhR and analogues reported. Sensitivity is 46.88 pg/mL with a detection range of 78.13-5000 pg/mL, total assay time of 4.5 hours, and a 100 µL sample volume. Reproducibility is reported with both intra-assay and inter-assay coefficients of variation below 15%. The kit is supplied with a CLIA plate, reference standard, concentrated biotinylated detection antibody (100×), concentrated HRP conjugate (100×), reference standard and sample diluent, biotinylated detection antibody diluent, HRP conjugate diluent, concentrated wash buffer (25×), substrate reagents A and B, desiccant, plate sealer, and product documentation. Storage conditions vary by component, including -20°C for the CLIA plate, reference standard, detection antibody, and HRP conjugate (the latter two protected from light), and 4°C for the diluents and substrate reagents.

Recovery has been evaluated in serum, EDTA plasma, and cell culture media, with average recovery values of 104%, 107%, and 105% respectively. Linearity has been demonstrated across dilution factors from 1:2 to 1:16 in serum, EDTA plasma, and cell culture media. The kit supports cell biology research, providing a tool for the accurate detection of aryl hydrocarbon receptor levels in human biological samples.
