Aggrecan (ACAN) is a large aggregating chondroitin sulfate proteoglycan that forms massive complexes with hyaluronic acid in articular cartilage extracellular matrix, and quantifying its circulating fragments is widely used as a biomarker of cartilage degradation in osteoarthritis and joint-injury research. This Assay Genie quantitative immunoassay measures human aggrecan in research samples using a pre-coated microplate, biotinylated detection antibody, and HRP-streptavidin conjugate workflow. Common applications include monitoring proteoglycan release from IL-1β- or TNF-α-stimulated chondrocyte monolayer and pellet cultures, profiling cartilage turnover in serum from subjects with knee osteoarthritis, and characterizing aggrecan catabolism in intervertebral disc, synovial fluid, and meniscal tissue studies, including detection of ARGS-aggrecan neoepitope fragments released by ADAMTS-4 and ADAMTS-5 cleavage. All components should be refrigerated at 4 degrees C between uses and protected from light until first opening, and the assay is intended to detect both intact aggrecan and proteolytic fragments generated during cartilage matrix breakdown.
