Human Aggrecan ELISA Kit

Designed by Assay Genie for quantitative measurement of aggrecan, this Sandwich ELISA kit employs a double-antibody format with colorimetric readout. The assay is validated for reactivity with human samples and is listed for cell biology research. Aggrecan (gene symbols include ACAN and AGCAN; aliases include Aggrecan core protein, CSPCP, AGC1, CSPG1, MSK16, CSPGCP and SEDK) is a key extracellular matrix component of cartilage, and altered levels have been linked to joint disorders such as osteoarthritis.

Reported sensitivity is 0.469 ng/mL and the working range is 0.781–50 ng/mL. Compatible sample types include serum, plasma, cell culture supernatant, cell or tissue lysate, and other liquid samples. Reagents are stored at 2–8°C for 12 months. The procedure uses a biotin-labelled detection antibody, an HRP-Streptavidin (SABC) conjugate, and TMB substrate, with absorbance read at 450 nm after a stop step. Sample preparation notes describe serum collection (clot, 1000 × g, 20 min), plasma collection (anticoagulant tubes, 1000 × g, 15 min at 2–8°C), tissue homogenisation in PBS with protease inhibitors, cell culture supernatant clarification (2500 rpm, 5 min), and cell lysis using lysis buffer with protease inhibitors.

Reported linearity at 1:2, 1:4 and 1:8 dilutions (n = 5) is 88–97%, 94–101% and 90–105% for serum; 90–101%, 89–100% and 86–101% for EDTA plasma; and 83–98%, 84–96% and 87–99% for heparin plasma. Spike-and-recovery averages are 99% for serum (range 87–105%), 95% for EDTA plasma (88–105%) and 91% for heparin plasma (87–99%).
