The kit employs a sandwich chemiluminescence immunoassay (CLIA) principle, with a microplate pre-coated with an antibody specific to the target protein. Standards or samples bind to the immobilized antibody, followed by a biotinylated detection antibody and HRP-conjugated streptavidin to form a sandwich complex. After washing, a chemiluminescent substrate is added; the HRP enzyme catalyzes a light-emitting reaction whose intensity is directly proportional to the concentration of the target protein and is measured using a luminometer. The reported sensitivity is 37.5 pg/mL, with a detection range of 62.5-4000 pg/mL and a recovery of 80%-120%. The assay is completed in 3 h 30 min.

The kit recognizes Human AGC in samples, with no significant cross-reactivity or interference between Human AGC and analogues observed.
