Through direct binding to P-TEFb and ELL family proteins, AFF4 coordinates the release of paused RNA polymerase II and the productive elongation of cellular and viral genes. In HIV-1 biology, AFF4 serves as an essential cofactor for Tat-mediated transactivation of the integrated proviral genome. Chromosomal rearrangements involving AFF4 also contribute to mixed lineage leukemia (MLL)-rearranged oncogenesis, making it a target of interest in hematological cancer research.
