Researchers investigating mixed-lineage leukemia, MLL-AFF1 (MLL-AF4) fusion biology, or transcription-factor signaling can use the Assay Genie Human AFF1 (AF4/FMR2 Family, Member 1) ELISA Kit as a quantitative immunoassay for measuring endogenous AFF1 protein in human-derived samples. AFF1 is a nuclear transcription factor best known for its contribution to the MLL-AFF1 fusion gene generated by the t(4;11)(q21;q23) translocation, a recurrent chromosomal rearrangement implicated in infant and adult acute lymphoblastic leukemia. Laboratories studying chromatin regulation by the DOT1L/H3K79 axis, transcription factor complexes, or MLL-rearranged leukemias often need to monitor AFF1 protein levels in cell lysates and tissue homogenates to complement transcript-based and cytogenetic workflows.

The kit ships in a 96-assay plate configuration that produces a measurable color change readable on standard microplate instruments. Intended specimens include tissue homogenates and cell lysates, supporting leukemia cell-line models and primary-sample research where protein-level confirmation of AFF1 expression is needed. Reagents are intended for in vitro research use only and should be handled according to the package insert; user-prepared samples require dilution into the supplied assay buffer before loading.

For reliable performance, assay buffer and standards should be equilibrated to room temperature before use, unused strips returned to the sealed pouch with desiccant, and the substrate solution protected from light. Researchers running longitudinal studies can aliquot working reagents to limit freeze-thaw cycles. The kit ships with sufficient wells for up to 96 assays, including standard-curve points run in duplicate and quality controls for accurate interpolation of unknown samples. Pairing this ELISA with qPCR or Western blot on matched lysates gives a more complete view of AFF1 regulation, since protein abundance and mRNA do not always track one-to-one.
