Human Adrenomedullin ELISA Kit

The Sandwich ELISA from Assay Genie is designed for the precise measurement of adrenomedullin levels in human samples, including serum, plasma, cell culture supernatants, cell or tissue lysates, and other liquid samples. Using a colorimetric detection method with a double antibody sandwich format, the kit offers a sensitivity of 3.75 pg/mL and a standard range of 6.25-400 pg/mL.

Adrenomedullin is a key regulator of blood vessel function, playing a crucial role in cardiovascular health and homeostasis. It is also involved in physiological processes such as inflammation, vasodilation, and cell growth, with abnormal levels linked to hypertension, heart failure, and sepsis. Reactivity is human, with the target known by the synonyms ADM and AM (Uniprot P35318), and the kit is positioned for neuroscience research. Storage is 2-8°C for 12 months.

The procedure involves equilibrating reagents and TMB substrate to room temperature, loading standards and samples on the pre-coated plate, incubating at 37°C for 90 minutes for antigen binding, adding biotin-labeled detection antibody for 60 minutes, adding HRP-Streptavidin (SABC) for 30 minutes, developing color with TMB substrate in the dark for 10-20 minutes, and reading absorbance at 450 nm. Linearity testing across serum, EDTA plasma and heparin plasma (n = 5) at 1:2, 1:4 and 1:8 dilutions showed recoveries spanning 82-103%, while spike-recovery experiments gave average values of 94% (serum), 91% (EDTA plasma) and 97% (heparin plasma).
