Description

A sandwich ELISA from Assay Genie designed for the quantitative measurement of human ADP-ribosyl cyclase 2 (BST1) in serum, plasma, tissue homogenates, cell culture supernates and other biological fluids. The assay uses a colorimetric detection read-out and recognises the natural and recombinant forms of human ADP-ribosyl cyclase/cyclic ADP-ribose hydrolase 2.

BST1 (also known as Bone marrow stromal antigen 1, Cyclic ADP-ribose hydrolase 2, BST-1, cADPr hydrolase 2 and CD157) is a GPI-anchored cell membrane protein that functions as a homodimer. It synthesises the second messengers cyclic ADP-ribose and nicotinate-adenine dinucleotide phosphate, the former acting to release calcium from intracellular stores, and may be involved in pre-B-cell growth. Dysregulation of BST1 has been implicated in autoimmune disorders, inflammatory conditions and metabolic disorders, and its expression is enhanced in bone marrow stromal cell lines derived from patients with rheumatoid arthritis. The protein is catalogued under UniProt entry Q10588 with a molecular weight of 29,326 Da.

Performance characteristics include a detection range of 0.156-10 ng/mL and a sensitivity of 0.10 ng/mL, with an intra-assay CV of 3.9% and an inter-assay CV of 6.5%. Linearity has been demonstrated in serum (108-119%, 85-97%, 109-121% and 84-93% at 1:2, 1:4, 1:8 and 1:16 dilutions), EDTA plasma (106-117%, 100-110%, 82-92% and 105-114%) and heparin plasma (102-112%, 101-111%, 99-109% and 106-116%). Recovery averages 112% (range 106-118%) for serum and 114% (range 108-120%) for plasma. The kit is supplied with a dismountable microplate, lyophilised standard, sample and assay diluents, detection reagents A and B, wash buffer, substrate, stop solution and plate sealer, and is intended for research use only within the Development Biology research area.
