Human ADAMTS8 ELISA Kit

A colorimetric, double-antibody sandwich ELISA from Assay Genie enables quantitative detection of human ADAMTS8 (A disintegrin and metalloproteinase with thrombospondin motifs 8). The assay is suited to serum, plasma, cell culture supernatant, cell or tissue lysate, and other liquid samples, and the kit is stored at 2-8°C for 12 months.

Reported performance includes a sensitivity of 9.375 pg/mL and a working range of 15.625-1000 pg/mL. Linearity was demonstrated at 1:2, 1:4 and 1:8 dilutions: serum 85-100%, 86-103%, 85-100%; EDTA plasma 87-98%, 86-99%, 85-98%; heparin plasma 85-98%, 91-99%, 84-99%. Recovery ranges and averages were serum 88-103% (96%), EDTA plasma 85-103% (95%), and heparin plasma 86-102% (93%).

The workflow covers reagent and TMB substrate equilibration, primary incubation at 37°C for 90 minutes, biotin-labelled detection antibody for 60 minutes, HRP-Streptavidin (SABC) for 30 minutes, TMB colour development in the dark for 10-20 minutes, and absorbance measurement at 450 nm. ADAMTS8 contributes to extracellular matrix degradation, cell migration and tissue remodelling, with dysregulation linked to cancer, inflammation and cardiovascular disorders (UniProt Q9UP79).
