Human ADAM29 ELISA Kit

A colorimetric sandwich ELISA from Assay Genie configured as a double-antibody immunoassay for the quantitative measurement of human ADAM29. The kit is validated for use with human serum, plasma, cell culture supernatant, cell or tissue lysate, and other liquid sample types, and the protocol uses a biotin-labelled detection antibody with HRP-streptavidin and TMB substrate, read at 450 nm after addition of stop solution.

Linearity was assessed in serum, EDTA plasma and heparin plasma at 1:2, 1:4 and 1:8 dilutions, with recovery ranges of 87–98%, 85–101% and 91–99% for serum; 83–100%, 91–98% and 82–95% for EDTA plasma; and 82–98%, 82–90% and 86–100% for heparin plasma. Spike-and-recovery experiments gave average recoveries of 93% in serum, 98% in EDTA plasma and 93% in heparin plasma. Unopened components are stored at 2–8°C for 12 months.

ADAM29 is a disintegrin and metalloproteinase domain-containing protein implicated in cell adhesion and migration, with established roles in embryogenesis and tissue remodelling, and its dysregulation has been linked to cancer and inflammatory disorders. The supplied microplate is pre-coated and the workflow includes primary incubation at 37°C for 90 minutes, detection antibody binding for 60 minutes, and HRP-streptavidin binding for 30 minutes, with TMB colour development in the dark for 10–20 minutes.
