Human ADAM10 ELISA Kit

From Assay Genie, this sandwich ELISA kit is designed for the quantitative detection of human ADAM10 (a disintegrin and metalloproteinase domain-containing protein 10), a key enzyme involved in the shedding of cell surface proteins and implicated in cancer, Alzheimer's disease, and inflammatory disorders. The double-antibody assay uses colorimetric detection and is suited to signal transduction research.

Performance specifications include a sensitivity of 37.5 pg/mL and an assay range of 62.5-4000 pg/mL. Compatible sample types include serum, plasma, cell culture supernatant, cell or tissue lysate, and other liquid samples. Linearity data (n=5) at 1:2, 1:4, and 1:8 dilutions: serum 86-102%, 93-105%, 85-105%; EDTA plasma 86-98%, 90-99%, 86-101%; heparin plasma 82-98%, 81-98%, 81-99%. Recovery testing (n=5) yielded averages of 91% for serum (range 86-96%), 95% for EDTA plasma (range 90-103%), and 95% for heparin plasma (range 89-105%).

The protocol uses a pre-coated plate with incubations at 37°C: 90 minutes for antigen binding, 60 minutes with biotin-labeled detection antibody, and 30 minutes with HRP-Streptavidin (SABC), followed by TMB substrate development in the dark for 10-20 minutes and absorbance measurement at 450 nm after adding stop solution. The kit provides a dismountable ELISA microplate, lyophilized standard, biotin-labeled antibody and HRP-Streptavidin conjugate (each 100X concentrate), TMB substrate, sample/antibody/SABC dilution buffers, stop solution, 25X wash buffer, plate sealers, and a technical manual. Kits are stored at 2-8°C for 12 months.
