Description

From Assay Genie, this CLIA kit is designed for the in vitro quantitative determination of Human ADAM10 (A Disintegrin And Metalloprotease 10) in serum, plasma and other biological fluids. The assay uses a Sandwich-CLIA principle: the microplate is pre-coated with an antibody specific to Human ADAM10, samples or standards are added, followed by a biotinylated detection antibody and an Avidin-HRP conjugate, with signal developed using a chemiluminescent substrate and read as Relative Light Units (RLU) on a chemiluminescence immunoassay analyzer. The documented assay time is 4.5h with a sample volume of 100 μL, a stated sensitivity of 9.38 pg/mL, and a detection range of 15.63–1000 pg/mL. The kit is reactive with Human samples, and according to the manufacturer no significant cross-reactivity or interference between Human ADAM10 and analogues was observed.

Supplied kit components include a pre-coated CLIA plate, reference standard, concentrated biotinylated detection antibody (100×), concentrated HRP conjugate (100×), reference standard and sample diluent, biotinylated detection antibody diluent, HRP conjugate diluent, concentrated wash buffer (25×), substrate reagents A and B, desiccant, plate sealer and a product description.

In the manufacturer's linearity evaluation, dilution ranges in serum (n=5) were 102–119% (1:2), 100–115% (1:4), 96–111% (1:8) and 87–97% (1:16); in EDTA plasma (n=5) the corresponding ranges were 84–98%, 103–117%, 87–100% and 85–99%; and in cell culture media (n=5) ranges were 99–111%, 101–115%, 96–109% and 96–109%. Spiked recovery averaged 101% for serum (range 95–106%), 97% for EDTA plasma (range 91–107%) and 97% for cell culture media (range 92–106%). Stated reproducibility specifies intra-CV and inter-CV less than 15%, with intra-assay CVs of 12.27%, 9.96% and 9.04% across three samples and inter-assay CVs of 11.58%, 11.64% and 6.1%.
