Description

From Assay Genie, this competitive ELISA kit is designed for the quantitative measurement of adalimumab in human serum, plasma, cell culture supernatant, cell or tissue lysate, and other liquid samples. Adalimumab is a therapeutic monoclonal antibody used to treat autoimmune conditions such as rheumatoid arthritis, psoriasis, and Crohn's disease, and monitoring its levels in biological samples supports research into treatment optimisation and patient outcomes.

The assay uses a competitive ELISA format coated with antigen, with colourimetric detection. It is validated for human samples, with a stated sensitivity of 0.188 ng/mL and a working range of 0.313–20 ng/mL. Linearity at 1:2, 1:4, and 1:8 dilutions has been established in serum (n=10) at 90–102%, 82–99%, and 80–98%; EDTA plasma (n=10) at 85–104%, 91–94%, and 84–97%; and heparin plasma (n=10) at 91–102%, 89–99%, and 80–96%. Recovery in serum (n=10) ranges from 92–99% with an average of 96%; EDTA plasma (n=10) ranges from 91–101% with an average of 98%; and heparin plasma (n=10) ranges from 87–102% with an average of 94%.

The protocol begins with equilibration of the TMB substrate for 30 minutes at room temperature, followed by adding 50 µL of standard or sample with 50 µL of biotin-labelled antibody per well and incubating at 37°C for 45 minutes. The plate is washed five times, TMB substrate is added, and the plate is incubated in the dark at 37°C for 10–20 minutes. After stopping the reaction, the colour changes from blue to yellow, and absorbance is read at 450 nm. The kit should be stored at 2–8°C in sealed conditions; cryopreservation is not recommended.
