Description

From Assay Genie, this sandwich ELISA kit uses a double-antibody format with colorimetric detection to quantify ACTN2/Sarcomeric Alpha Actinin in human samples. The kit is intended for serum, plasma, cell culture supernatant, cell or tissue lysates, and other liquid samples.

Reported performance figures include a sensitivity of 0.188 ng/mL and a measuring range of 0.313–20 ng/mL. Linearity was evaluated in serum, EDTA plasma, and heparin plasma at 1:2, 1:4, and 1:8 dilutions. In serum, the percentages ranged 86–105% at 1:2, 87–103% at 1:4, and 85–104% at 1:8. EDTA plasma yielded 83–96%, 84–101%, and 84–101% at the same dilutions, while heparin plasma yielded 82–95%, 81–90%, and 82–94%. Spike-and-recovery testing showed averages of 96% in serum (range 88–104%), 95% in EDTA plasma (88–99%), and 95% in heparin plasma (89–101%).

The procedure begins with reagent equilibration to room temperature, followed by primary incubation at 37°C for 90 minutes, biotin-labelled detection antibody binding at 37°C for 60 minutes, and HRP-streptavidin binding at 37°C for 30 minutes. TMB substrate is added and developed in the dark for 10–20 minutes, after which stop solution is introduced and absorbance is read at 450 nm. Unopened kits are stored at 2–8°C for up to 12 months. The target corresponds to UniProt entry P35609, with listed synonyms ACTN2, Actinin Alpha 2, and CMD1AA, and the kit falls within the Cell Biology research area.
