Description

From Assay Genie, this colorimetric sandwich ELISA is designed for the quantitative detection of human ACTA1 (alpha skeletal muscle actin) in research samples. The kit uses a double-antibody sandwich format and is validated for use with human serum, plasma, cell culture supernatant, cell or tissue lysate, and other liquid sample types.

Reported analytical performance includes a sensitivity of 0.188 ng/mL and a working range of 0.313–20 ng/mL. Linearity has been assessed at 1:2, 1:4 and 1:8 dilutions in serum (88–103%, 86–105%, 90–102%), EDTA plasma (83–98%, 93–99%, 86–97%) and heparin plasma (81–96%, 85–96%, 84–100%), with average spike-and-recovery values of 96% for serum and 94% for both EDTA and heparin plasma. The target protein corresponds to UniProt P68133 and is described as playing a role in muscle contraction, with dysregulation linked to various muscle disorders, making it a biomarker of interest in muscle physiology and related disease research. Reagents are stored at 2–8°C for up to 12 months, with selected components suitable for longer-term storage at –20°C as described in the supplied manual.

The workflow follows a standard sandwich ELISA procedure: equilibrate reagents and TMB substrate, load standards, samples and controls onto the pre-coated plate and incubate at 37°C for 90 minutes for antigen binding; add biotin-labelled detection antibody and incubate at 37°C for 60 minutes; add HRP-Streptavidin (SABC) and incubate at 37°C for 30 minutes; develop with TMB substrate in the dark for 10–20 minutes; then add stop solution and read absorbance at 450 nm.
