The Human ACSL4/FACL4 ELISA Kit from Assay Genie enables quantitative measurement of long-chain acyl-CoA synthetase 4, an enzyme positioned at the crossroads of lipid metabolism and ferroptosis. ACSL4 preferentially activates polyunsaturated fatty acids such as arachidonic acid, esterifying them into phospholipid pools that become substrates of lipid peroxidation during iron-dependent cell death. Researchers studying hepatocellular carcinoma, triple-negative breast cancer, prostate cancer, and other malignancies where ACSL4 expression predicts ferroptosis sensitivity use this assay to stratify cell lines and patient specimens. Beyond oncology, the kit supports investigations into X-linked intellectual disability linked to FACL4 loss-of-function variants on Xq23, metabolic syndrome, and non-alcoholic fatty liver disease.

The assay uses a quantitative immunometric configuration with all reagents supplied ready-to-use. Standard ELISA workflow is followed, with absorbance read on a standard microplate reader. Each microplate provides 96 determinations, with sufficient reagents for running standard curves in duplicate. Components include a pre-coated plate, biotinylated detection antibody, streptavidin-HRP conjugate, calibrated recombinant standards, wash buffer, and substrate solution. The assay is intended for research use only and is not validated for diagnostic procedures. Human-specific capture and detection antibodies minimize cross-reactivity with rodent or non-human primate homologs, supporting clean discrimination of endogenous human antigen.

Sample handling guidance: collect blood-derived specimens using standard venipuncture techniques, aliquot to avoid repeated freeze-thaw cycles, and store at -80°C for long-term preservation. Allow samples to equilibrate to room temperature before loading. This Assay Genie product supports studies aiming to correlate circulating ACSL4 levels with disease stage, treatment response, or ferroptosis induction in preclinical models.
