Quantifying the cytoplasmic bifunctional enzyme iron regulatory protein 1 (IRP1), also annotated as IREB1, IREBP, cytoplasmic aconitate hydratase, citrate hydro-lyase, and ferritin repressor protein (UniProt P21399), this Assay Genie ELISA Kit pairs with a 96 assays microplate workflow. The kit targets human Aconitase 1 (ACO1), an iron–sulfur cluster protein that interconverts citrate and isocitrate in the cytosol and, upon iron depletion, loses its cluster to bind iron responsive elements and post-transcriptionally regulate ferritin and transferrin receptor mRNAs. Because of this dual metabolic and RNA-binding role, ACO1 quantification supports research on iron homeostasis disorders, neurodegeneration, mitochondrial dysfunction, metabolic disease, and inflammation.

The detection range is 0.312 to 20 ng/mL, generated from serial dilutions of the included calibrator (20, 10, 5, 2.5, 1.25, 0.625, 0.312, and 0 ng/mL) across an 8-point standard curve. Sensitivity of this assay is 0.115 ng/mL, with the lower limit of detection determined statistically as two standard deviations above the mean of twenty zero-standard replicates. The kit shows no significant cross-reactivity or interference between human ACO1 and analogues, and is intended for in vitro research use only.

Each pack contains a precoated microplate, recombinant human ACO1 calibrator, biotinylated detection antibody, streptavidin-HRP conjugate, wash buffer, substrate, and stop solution. Stored refrigerated between uses with a shelf life of 12 months, the components remain stable; allow all reagents to equilibrate to room temperature before running the assay and protect the substrate from light.
