Human AChR/Anti-Acetylcholine Receptor ELISA Kit

Assay Genie offers a double antigen sandwich ELISA for the quantitative measurement of anti-acetylcholine receptor (AChR) antibodies in human samples. Anti-acetylcholine receptor antibodies are associated with autoimmune conditions such as myasthenia gravis, where they target acetylcholine receptors at the neuromuscular junction, and the kit is intended to support research in autoimmune disease, neurology and immunology.

Reactivity is human and the assay is compatible with serum, plasma, cell culture supernatant, cell or tissue lysate, and other liquid samples. Reported sensitivity is 0.938 ng/mL with a measuring range of 1.563–100 ng/mL. Sealed components are stored at 2–8 °C and should not be cryopreserved. The procedure uses a 90-minute primary incubation at 37 °C, a 60-minute biotin-labeled antigen binding step, a 30-minute HRP-streptavidin incubation, TMB color development in the dark for 10–20 minutes, and absorbance read at 450 nm after the stop solution is added.

Validation figures supplied by the manufacturer cover serum, EDTA plasma and heparin plasma. Linearity across dilutions of 1:2, 1:4 and 1:8 ranges from 82–100%, 84–95% and 92–103% for serum; 83–100%, 84–92% and 88–96% for EDTA plasma; and 82–100%, 80–95% and 89–101% for heparin plasma (n = 5 for each). Recovery ranges are 85–98% (serum, average 90%), 85–98% (EDTA plasma, average 95%) and 86–99% (heparin plasma, average 94%).
