Description

Assay Genie's sandwich ELISA kit is designed for the quantitative detection of the human ABO histo-blood group system transferase enzyme in biological samples. The enzyme underpins ABO blood group determination and contributes to immune response and tissue development, with dysregulation reported across multiple disease contexts. The kit is suited to applications in immunology, genetics, and clinical pathology research.

The assay uses a double-antibody sandwich ELISA format with colorimetric readout. Reactivity is limited to human samples, and validated sample types include serum, plasma, cell culture supernatant, cell or tissue lysate, and other liquid samples. The kit is stored at 2–8°C for 12 months. Reported linearity: serum (n=5) 86–96% at 1:2, 88–97% at 1:4, 84–97% at 1:8; EDTA plasma (n=5) 88–105% at 1:2, 89–99% at 1:4, 82–100% at 1:8; heparin plasma (n=5) 88–102% at 1:2, 83–98% at 1:4, 85–96% at 1:8. Recovery: serum (n=5) range 85–100%, average 92%; EDTA plasma (n=5) range 87–104%, average 94%; heparin plasma (n=5) range 89–102%, average 95%.

The protocol equilibrates reagents and TMB substrate to room temperature, loads standards, samples and blanks on the pre-coated plate, and incubates at 37°C for 90 minutes for antigen binding. A biotin-labeled detection antibody is added and incubated at 37°C for 60 minutes, followed by HRP-Streptavidin (SABC) for 30 minutes. TMB substrate is incubated in the dark for 10–20 minutes; stop solution is then added and absorbance is read at 450 nm.
