ABHD12 (abhydrolase domain-containing 12) functions as a lysosomal monoacylglycerol lipase that hydrolyzes 2-arachidonoylglycerol and related endocannabinoid substrates. Loss-of-function mutations in the gene are linked to PHARC syndrome, a rare neurological disorder combining polyneuropathy, hearing loss, ataxia, retinitis pigmentosa, and early-onset cataract. The protein is highly expressed in microglia and macrophages of the central nervous system, where it regulates endocannabinoid tone and inflammatory signaling.
