Human ABCC6/MRP6 ELISA Kit

From Assay Genie, this Sandwich ELISA Kit is designed for the quantitative measurement of ABCC6 (also known as Multidrug Resistance-Associated Protein 6, or MRP6) in human samples. The kit is reactive with human samples and accepts serum, plasma, cell culture supernatant, cell or tissue lysate, and other liquid samples. Detection is colorimetric, performed on a dismountable pre-coated microplate with absorbance read at 450 nm after TMB color development and stop solution.

The assay uses a sandwich ELISA, double-antibody format with a biotin-labeled detection antibody and HRP-Streptavidin conjugate (SABC). The reported sensitivity is 46.875 pg/mL and the assay range is 78.125–5000 pg/mL. Kit components include the ELISA microplate, lyophilized standard, biotin-labeled antibody (100X), HRP-Streptavidin conjugate SABC (100X), TMB substrate, sample dilution buffer, antibody dilution buffer, SABC dilution buffer, stop solution, 25X wash buffer, plate sealer and a technical manual. Unopened components are stored at 2-8°C for 12 months, with certain items additionally suitable for 12 months at -20°C.

ABCC6 is a transporter protein involved in moving molecules across cell membranes, and its dysfunction has been linked to pseudoxanthoma elasticum (PXE), a rare genetic disorder characterized by mineralization of elastic fibers in tissues. Linearity in serum (n=5) ranged from 87-105% at 1:2, 85-105% at 1:4 and 92-104% at 1:8, with EDTA plasma at 86-99%, 83-99% and 87-100% and heparin plasma at 89-100%, 85-98% and 83-99%. Recovery averaged 93% in serum (range 85-104%), 97% in EDTA plasma (range 93-99%) and 97% in heparin plasma (range 86-103%).
