The Assay Genie ADAM15 immunoassay is configured as a sandwich ELISA in which endogenous human ADAM15 captured from serum, plasma, or cleared tissue homogenate is bound by the pre-coated polyclonal capture antibody, then detected with a biotinylated polyclonal reporter developed via streptavidin-HRP and TMB substrate; absorbance is read at 450 nm against a reference wavelength and back-calculated against the reconstituted standard curve. The capture and detection antibodies were raised against recombinant human ADAM15 extracellular region and show no significant cross-reactivity with homologous family members ADAM9, ADAM10, ADAM12, or ADAM17 under standard run conditions, supporting clean quantitation in mixed biological matrices. Cell culture supernatant and cell lysate samples can be assayed directly after centrifugation at 1000 x g for 10 minutes to remove debris, while EDTA plasma should be collected with minimal hemolysis to avoid falsely elevated readings. A typical run from sample loading to final plate read takes approximately 2.5 to 3 hours at room temperature, after which stop solution is added and absorbance recorded within 10 minutes. Reported intra-assay CV is below 8% and inter-assay CV below 10% across the working range, with recovery of spiked recombinant ADAM15 in human serum consistently in the 85 to 115% window.
