Description

Assay Genie provides a sandwich ELISA kit for the quantitative measurement of human 78 kDa glucose-regulated protein (HSPA5), also referred to as BiP, GRP-78, GRP78, Binding-immunoglobulin protein, and Immunoglobulin heavy chain-binding protein. The kit is validated for reactivity with human samples and is suitable for serum, plasma, tissue homogenates, cell culture supernates and other biological fluids. Detection is performed by colorimetric ELISA in a solid phase format.

Performance characteristics from the manufacturer include a detection range of 15.6–1000 pg/mL and a sensitivity of 5.6 pg/mL. Intra-assay precision is 4.2% CV and inter-assay precision is 7.9% CV. Linearity was evaluated in serum, EDTA plasma and heparin plasma at dilutions of 1:2, 1:4, 1:8 and 1:16, with reported ranges of 93–120% for serum, 85–114% for EDTA plasma and 86–123% for heparin plasma across the dilutions tested. Average recovery is 84% (range 80–90%) in serum and 86% (range 80–92%) in plasma. The kit specifically detects natural and recombinant human endoplasmic reticulum chaperone BiP (UniProt P11021).

HSPA5/BiP is an endoplasmic reticulum chaperone with a central role in protein folding and quality control, and its dysregulation has been associated with cancer, diabetes and neurodegenerative disorders. The product is supplied for research use only, with shipping at +4°C and storage at –20°C. The associated research area is neurosciences, and the target protein localises to the endoplasmic reticulum lumen, melanosome and cytoplasm.
