Description

Assay Genie provides a Sandwich ELISA Kit designed for the quantitative measurement of human 7,8-dihydro-8-oxoguanine triphosphatase (NUDT1), also known as MTH1. NUDT1 is an antimutagenic enzyme that acts as a sanitising enzyme for oxidised nucleotide pools, hydrolysing 8-oxo-dGTP, 8-oxo-dATP and 2-OH-dATP to prevent misincorporation of oxidised purine nucleoside triphosphates into DNA and thereby suppress A:T to C:G and G:C to T:A transversions. The assay is configured for human samples with a reported detection range of 0.78–50 ng/mL and a sensitivity of 0.23 ng/mL.

The kit is validated for use with serum, plasma, tissue homogenates, cell culture supernatants and other biological fluids. Reported precision figures are an intra-assay CV of 6.1% and an inter-assay CV of 8.5%. Recovery is 86% (80–92%) in serum and 88% (82–94%) in plasma, and linearity has been demonstrated across serial dilutions of serum, EDTA plasma and heparin plasma. Specificity is reported for natural and recombinant human 7,8-dihydro-8-oxoguanine triphosphatase.

According to UniProt entry P36639, NUDT1 functions as a monomer, with isoform p26 localised to the cytoplasm and the mitochondrial matrix. The protein is implicated in DNA repair, dGTP and dATP catabolic processes, the purine nucleotide catabolic process, and the response to oxidative stress. The kit is supplied for research use only.
