Description

Assay Genie's Sandwich ELISA Kit is designed for the accurate quantification of human 6-phosphogluconate dehydrogenase, decarboxylating (PGD) levels in serum, plasma, tissue homogenates, cell culture supernates and other biological fluids. The kit demonstrates reactivity with human samples and is suited to research into the pentose phosphate pathway, where PGD catalyses the oxidative decarboxylation of 6-phosphogluconate to ribulose 5-phosphate and CO2, with concomitant reduction of NADP to NADPH.

With a detection range of 1.56-100 umol/L and a sensitivity of 0.78 umol/L, the kit offers reproducible quantification. Intra-assay precision is reported at 4.8% CV and inter-assay precision at 6.1% CV. Linearity has been validated across serum (99-108%, 97-107%, 101-111%, 85-95%), EDTA plasma (102-112%, 90-101%, 103-114%, 114-124%) and heparin plasma (109-120%, 88-97%, 90-99%, 109-119%) at dilutions of 1:2, 1:4, 1:8 and 1:16. Recovery is 93% (range 87-99%) for serum and 95% (range 89-101%) for plasma.

The assay recognises natural and recombinant human PGD (synonym: PGDH), which functions as a homodimer localised to the cytoplasm, with UniProt accession P52209 and a molecular weight of 51,872 Da. PGD is classified under EC 1.1.1.44, and its dysregulation has been associated with cancer, metabolic disorders and neurodegenerative diseases. For research use only.
