CD73, also called ecto-5'-nucleotidase (NT5E), is a GPI-anchored cell-surface ectoenzyme that converts extracellular adenosine monophosphate into adenosine and inorganic phosphate. This enzymatic activity drives immunosuppressive adenosine signaling through A2A receptors on T cells, positioning CD73 as a focal target in tumor immune evasion studies and an emerging biomarker for predicting response to checkpoint blockade therapy. This Assay Genie human CD73 immunoassay supports quantitative measurement of the enzyme in cell culture supernatant, human plasma samples, and serum specimens, making it a practical choice for oncology, immunology, and adenosine-pathway research programs.

Configured for use with a pre-coated microplate, the kit supplies a matched pair of capture and detection antibodies, a recombinant human 5'-Nucleotidase standard calibrated for the assay, streptavidin-enzyme conjugate, substrate solution, and wash buffer. The 96 assays configuration provides enough wells to run full standard curves alongside experimental samples, supporting both single-analyte and screening-style studies. Researchers investigating anti-CD73 antibody therapeutics, adenosine receptor pharmacology, or immune cell exhaustion in the tumor microenvironment can integrate the kit into standard immunoassay workflows.

The procedure uses an HRP-TMB colorimetric readout with absorbance proportional to CD73 levels in each sample. Suitable applications include biomarker quantification, monitoring of CD73 shedding from tumor cells in vitro, and pharmacodynamic studies of small-molecule or biologic CD73 inhibitors. All components are intended for research use only.
