Description

This Assay Genie Sandwich ELISA Kit is designed for the quantitative measurement of human 5'-AMP-activated protein kinase subunit beta-1 (PRKAB1), also referred to as AMPK subunit beta-1 or AMPK. The assay is reactive with human samples and is intended for cardiovascular research, for research use only.

Reported performance characteristics include a detection range of 0.312–20ng/mL and a sensitivity of 0.16ng/mL. Intra-assay CV is 6.7% and inter-assay CV is 10.6%. Linearity in serum (N=5) ranges from 106–115% at 1:2, 84–97% at 1:4, 108–118% at 1:8, and 113–122% at 1:16. EDTA plasma (N=5) shows 92–102%, 107–117%, 108–116%, and 114–124% across the same dilutions, and heparin plasma (N=5) shows 109–120%, 102–111%, 94–105%, and 103–112%. Average recovery is 92% (range 86–98%) for serum and 94% (range 88–100%) for plasma. The kit is specific for natural and recombinant human PRKAB1.

PRKAB1 is the non-catalytic beta-1 subunit of the AMP-activated protein kinase (AMPK) heterotrimer, which also contains an alpha catalytic subunit (PRKAA1 or PRKAA2) and a gamma non-catalytic subunit (PRKAG1, PRKAG2 or PRKAG3). The beta subunit acts as a scaffold on which the AMPK complex assembles, bridging the alpha and gamma subunits. AMPK is an energy-sensing protein kinase that, in response to reduced intracellular ATP levels, activates energy-producing pathways and inhibits energy-consuming processes such as protein, carbohydrate and lipid biosynthesis, as well as cell growth and proliferation. The UniProt accession for the target is Q9Y478. Validated sample types include serum, plasma, tissue homogenates, cell culture supernates, and other biological fluids. Detection is colorimetric, with absorbance measured at 450 nm.
