Description

Manufactured by Assay Genie, this sandwich ELISA kit is designed for the quantitative measurement of human 3-hydroxyanthranilate 3,4-dioxygenase (HAAO) in biological samples. It is intended for research use only and shows reactivity with human samples, including serum, plasma, tissue homogenates, cell culture supernates and other biological fluids. HAAO, also known by the synonyms 3-hydroxyanthranilate oxygenase, 3-hydroxyanthranilic acid dioxygenase, 3-HAO, and HAD, is a monomeric cytosolic enzyme that plays a pivotal role in tryptophan metabolism through the kynurenine pathway. Dysregulation of HAAO has been implicated in inflammatory disorders and neurological conditions.

The kit delivers a detection range of 0.312-20 ng/mL with a sensitivity of 0.22 ng/mL. Precision performance is reported with an intra-assay CV of 4.8% and an inter-assay CV of 7.9%. Linearity has been evaluated in serum, EDTA plasma, and heparin plasma across dilutions of 1:2, 1:4, 1:8, and 1:16 (N=5), with reported ranges of 83-120%. Recovery testing shows an average of 99% (range 93-105%) for serum and 101% (range 95-107%) for plasma.

Functionally, HAAO catalyzes the oxidative ring opening of 3-hydroxyanthranilate to 2-amino-3-carboxymuconate semialdehyde, which spontaneously cyclizes to quinolinate. The enzyme has a molecular weight of 17,275 Da and is classified as an oxidoreductase (EC 1.13.11.6). Specificity is directed toward natural and recombinant human HAAO, and the cited research area is neurosciences.
