Human 15-LO (Arachidonate 15-Lipoxygenase) ELISA Kit (HUES01471)

Assay Genie provides a sandwich ELISA kit for the quantitative measurement of human Arachidonate 15-lipoxygenase (ALOX15), also referred to by the synonyms 15-LOX, 15-LOX-1, 15LOX-1, and LOX15 (UniProt P16050). The assay uses a colorimetric readout and is reactive with human samples, including serum, plasma, tissue homogenates, cell culture supernates and other biological fluids. Specificity covers natural and recombinant human Arachidonate 15-lipoxygenase.

Manufacturer specifications report a detection range of 0.156-10 ng/mL and a sensitivity of 0.085 ng/mL, with an intra-assay CV of 7.1% and an inter-assay CV of 9.2%. Linearity across 1:2 to 1:16 dilutions yielded 91-101% to 91-101% recovery in serum (N=5), 98-109% to 105-114% in EDTA plasma (N=5), and 115-125% to 87-97% in heparin plasma (N=5). Mean recovery was 99% (range 93-105%) for serum and 101% (range 95-107%) for plasma.

ALOX15 is a non-heme iron-containing dioxygenase that catalyses the stereospecific peroxidation of free and esterified polyunsaturated fatty acids, generating bioactive lipid mediators. It converts arachidonic acid into 12-HPETE and 15-HPETE, and linoleic acid to 13-hydroperoxyoctadecadienoic acid. The enzyme contributes to immune and inflammatory responses, clearance of apoptotic cells, regulation of actin polymerisation, the cellular response to IL13/interleukin-13, corneal epithelial wound healing, the endoplasmic reticulum stress response, and the regulation of bone mass. It is predominantly cytosolic, translocates to the plasma membrane upon IL13 stimulation, and localises to lipid droplets. Research applications include the cardiovascular area.
