Description

Assay Genie offers a cell-based ELISA kit for the sensitive and accurate detection of phosphorylated HSP27 (heat shock protein 27) at serine 15 in cell lysates. The kit uses colorimetric detection at 450 nm and is intended for use with human samples, with a stated dynamic range of > 5000 Cells. Designed as a lysate-free, high-throughput assay, it enables researchers to quantify relative levels of phosphorylated HSP27 in cultured cells and to screen for the effects of various treatments, inhibitors (such as siRNA or chemicals), or activators on HSP27 phosphorylation.

The assay follows an indirect ELISA format in which HSP27 (Phospho-Ser15)-specific primary antibodies capture the target, and HRP-conjugated secondary antibodies bind the Fc region of the primary antibody to catalyze a colorimetric reaction upon substrate addition. Because the assay is qualitative, two normalisation approaches are provided: a monoclonal antibody specific for human GAPDH serves as an internal positive control to normalise target absorbance values, and the Crystal Violet whole-cell staining method may be used to determine cell density so that absorbance values can be adjusted for plating differences.

The target is encoded by the HSPB1 gene (Gene ID: 3315; UniProt ID: P04792), and the kit is supplied as a phospho-specific Cell-Based ELISA for the immunology research area. Phosphorylation of HSP27 at serine 15 is a critical post-translational modification that regulates this chaperone protein's function in cellular stress responses, with dysregulation implicated in cancer, neurodegenerative disorders, and cardiovascular diseases.
