Description

From Assay Genie, this Cell-Based ELISA Kit is intended for the qualitative determination of HSP27 protein expression directly in cultured cells, using a colorimetric readout measured at 450 nm. The assay is described as lysate-free, high-throughput, and sensitive, and it uses an indirect ELISA format in which HSP27-specific primary antibodies are captured and detected by HRP-conjugated secondary antibodies that catalyze a colorimetric reaction upon substrate addition.

Reactivity is stated for Human, Mouse, and Rat, and the indicated sample type is adherent cells. The kit is designed for measuring the relative amounts of HSP27 in cultured cells and for screening the effects that various treatments, inhibitors (such as siRNA or chemicals), or activators have on HSP27. Two normalization strategies are built into the workflow: a monoclonal anti-GAPDH antibody is included to serve as an internal positive control in normalizing the target absorbance values, and a Crystal Violet whole-cell staining step can be used to normalize absorbance to cell density. Storage is at 4°C with a stated stability of 6 months, and the research area is listed as Immunology. The reported dynamic range is > 5000 Cells.

HSP27, also known as heat shock protein 27 (gene symbol HSPB1), is a small heat shock protein involved in cellular stress responses and in regulating cell survival and apoptosis. Abnormalities in HSP27 levels have been linked to cancer, neurodegenerative disorders, and cardiovascular diseases, and the kit is positioned as a tool for studying disease mechanisms and potential therapeutic targets.
