HSF2 Colorimetric Cell-Based ELISA

A Cell-Based ELISA from Assay Genie designed for the qualitative determination of HSF2 (Heat Shock Factor 2) in cultured cells, providing a convenient, lysate-free and high-throughput approach to measuring HSF2 protein expression profiles. The assay can be used to study the effects of various treatments, inhibitors such as siRNA or chemicals, and activators on HSF2 levels, and is suitable for adherent cells.

Detection relies on a colorimetric readout at 450 nm in an indirect ELISA format, in which HSF2-specific primary antibodies capture the target while HRP-conjugated secondary antibodies catalyse a colorimetric reaction upon substrate addition. A monoclonal anti-GAPDH antibody is included as an internal positive control for normalising target absorbance, and a Crystal Violet whole-cell staining step allows cell density to be determined so that absorbance values can be normalised to cell number. The kit is reactive with human, mouse and rat samples and has a stated dynamic range of > 5000 cells.

HSF2 is a DNA-binding protein of the heat shock factor family that binds heat shock promoter elements (HSE) and activates transcription in response to cellular stress, helping to maintain cellular homeostasis. Dysregulation of HSF2 has been implicated in cancer, neurodegenerative disorders and cardiovascular conditions, underlining its interest as a research target. The kit is supplied in a 96-well microplate format and is intended to be stored at 4°C with a stated stability of 6 months.
