HSF1 Colorimetric Cell-Based ELISA Kit

From Assay Genie, this Cell-Based ELISA Kit enables the qualitative measurement of Heat Shock Factor 1 (HSF1) protein expression directly in cultured adherent cells using a lysate-free, indirect ELISA format. The assay is reactive with human and mouse samples and uses a colorimetric detection method at 450 nm. It can be used to measure the relative amounts of HSF1 in cultured cells and to screen the effects of various treatments, inhibitors (such as siRNA or chemicals), or activators on HSF1.

The workflow captures HSF1 with target-specific primary antibodies, and HRP-conjugated secondary antibodies then catalyze a colorimetric reaction upon substrate addition. A monoclonal anti-human GAPDH antibody is included as an internal positive control for normalizing target absorbance values, and a Crystal Violet whole-cell staining step can be used to determine cell density, allowing results to be normalized to cell amounts and correct for plating differences.

The kit is supplied in a 96-well microplate format with a dynamic range of > 5000 Cells. It is stored at 4°C with a stated stability of 6 months and is intended for research in the Epigenetics and Nuclear Signaling area, supporting studies of stress response, cellular homeostasis, and disease mechanisms in which HSF1 dysregulation has been implicated, including cancer, neurodegenerative disorders, and metabolic conditions.
