Description

This Colorimetric Cell-Based ELISA Kit from Assay Genie is designed for the detection and quantification of HS1 phosphorylated at Tyr397. It is a lysate-free, high-throughput assay that enables measurement of relative HS1 phosphorylation levels in cultured adherent cells, making it suitable for screening the effects of various treatments, inhibitors (such as siRNA or chemicals), or activators on HS1 phosphorylation.

Detection uses an indirect ELISA format, in which HS1 (Phospho-Tyr397) is captured by phospho-specific primary antibodies, and HRP-conjugated secondary antibodies catalyze a colorimetric reaction measured at 450 nm. A monoclonal antibody specific for human GAPDH is included as an internal positive control for normalizing target absorbance, and Crystal Violet whole-cell staining is provided to determine cell density. The assay is reactive with human samples and has a dynamic range of > 5000 cells.

The target protein HS1, encoded by the gene symbol HCLS1 (Gene ID 3059, UniProt P14317), is described as a substrate of the antigen receptor-coupled tyrosine kinase that plays a role in antigen receptor signaling for both clonal expansion and deletion in lymphoid cells, with expression restricted to tissues and cells of hematopoietic origin.
