Description

From Assay Genie, this cell-based ELISA kit is designed to detect phosphorylation of HRS at tyrosine 334 in cellular samples. It is described as a convenient, lysate-free, high throughput and sensitive assay for measuring relative amounts of phosphorylated HRS in cultured cells, as well as for screening the effects of various treatments, inhibitors (such as siRNA or chemicals), or activators on HRS phosphorylation.

Detection is colorimetric at 450 nm, with reactivity against human, mouse, and rat samples and a dynamic range of > 5000 Cells. The kit uses a 96-well microplate format and an indirect ELISA design in which primary antibodies capture HRS (Phospho-Tyr334) and HRP-conjugated secondary antibodies catalyze a colorimetric reaction upon substrate addition. A monoclonal antibody specific for human GAPDH is included as an internal positive control for normalising target absorbance, and Crystal Violet whole-cell staining can be used to determine cell density after the HRP readout.

The target, HRS (gene symbol HGS; UniProt ID O14964; NCBI Gene ID 9146), is a zinc finger protein that is rapidly tyrosine phosphorylated in cells stimulated with various growth factors and regulates endosomal sorting as well as the recycling and degradation of membrane receptors. Phosphorylation at tyrosine 334 has been linked to cell growth, differentiation, and survival, with dysregulation of this event implicated in cancer, inflammation, and autoimmune disorders. The reported molecular weight is 76,376 Da.
