Description

From Assay Genie, this Cell-Based ELISA Kit is designed for the detection of HP1gamma levels in cell lysates and tissue extracts, providing a sensitive and specific tool for research applications. HP1gamma is a key protein involved in chromatin regulation and gene expression, with roles in cell division, DNA repair, and development. The kit supports studies into disease mechanisms associated with dysregulation of HP1gamma, including cancer, neurodegenerative disorders, and metabolic diseases.

The assay employs a colorimetric detection method read at 450 nm and is supplied in a 96-well microplate format suited to adherent cells. It demonstrates reactivity with human, mouse, and rat samples and supports a dynamic range of > 5000 Cells. Storage is at 4°C with 6 months stability. The kit uses an indirect ELISA format in which HP1gamma is captured by HP1gamma-specific primary antibodies, while HRP-conjugated secondary antibodies bind the Fc region of the primary antibody and catalyse a colour reaction upon substrate addition.

A monoclonal antibody specific for human GAPDH is included as an internal positive control for normalising target absorbance values, and a Crystal Violet whole-cell staining step can be used to determine cell density so that absorbance values can be adjusted to cell amounts. As a qualitative assay, the kit is suited to measuring relative HP1gamma levels in cultured cells and to screening the effects of treatments, inhibitors such as siRNA or chemicals, and activators on HP1gamma expression.
