Description

From Assay Genie, this cell-based ELISA kit is designed for detecting and quantifying HOXD12 protein levels in cell lysates, offering high sensitivity and specificity for a variety of research applications. The assay uses an indirect ELISA format in which HOXD12-specific primary antibodies are paired with HRP-conjugated secondary antibodies that catalyze a colorimetric reaction at 450 nm upon substrate addition. The workflow is lysate-free, high-throughput, and suited to screening the effects of treatments, inhibitors (such as siRNA or chemicals), or activators on HOXD12 expression in cultured cells.

The assay is configured for a 96-well microplate format and is reactive with human and mouse samples, with an expected dynamic range of greater than 5000 cells. Qualitative determination of HOXD12 concentration relies on normalization: a monoclonal anti-GAPDH antibody is included as an internal positive control, and an optional Crystal Violet whole-cell staining step allows absorbance values to be normalized to cell density, correcting for plating differences. Storage is at 4°C with a stated stability of 6 months, and the plate is read on a standard microplate reader capable of measuring absorbance at 450 nm.

HOXD12 is a sequence-specific transcription factor that is part of a developmental regulatory system providing positional identity along the anterior-posterior axis, with dysregulation implicated in developmental disorders and cancer. The kit is categorized under the Epigenetics and Nuclear Signaling research area.
