Description

Assay Genie's HOXB9 Colorimetric Cell-Based ELISA Kit is a cell-based assay designed for the measurement of HOXB9 levels in cell lysates and tissue samples. The kit employs a colorimetric detection method read at 450 nm and is supplied in a 96-well microplate format with a dynamic range of > 5000 cells. It is reactive with human and mouse samples and is intended for storage at 4°C for 6 months. The research area listed for this product is Epigenetics and Nuclear Signaling.

The assay uses an indirect ELISA format. HOXB9 is captured by HOXB9-specific primary antibodies, and HRP-conjugated secondary antibodies bind the Fc region of the primary antibody, catalysing a colorimetric reaction upon substrate addition. The kit is described as a convenient, lysate-free, high throughput and sensitive assay that can detect HOXB9 protein expression profile in cultured cells, and can be used to screen the effects of various treatments, inhibitors (such as siRNA or chemicals) or activators on HOXB9. Because the Cell-Based ELISA is qualitative, two normalisation methods are included: a monoclonal antibody specific for human GAPDH serves as an internal positive control for normalising target absorbance values, and Crystal Violet whole-cell staining may be used to determine cell density and adjust for plating differences.

HOXB9 is a sequence-specific transcription factor that is part of a developmental regulatory system providing cells with specific positional identities on the anterior-posterior axis, and it belongs to the Abd-B homeobox family. The encoded nuclear protein is involved in cell proliferation and differentiation, and increased expression has been associated with some cases of leukemia, prostate cancer and lung cancer.
