Description

Designed for the quantitative measurement of TNFα in equine samples, this Sandwich ELISA from Assay Genie is a sensitive and specific assay intended for veterinary research applications. TNFα is a key inflammatory cytokine involved in the regulation of immune responses and the development of inflammatory conditions in horses, making its monitoring valuable for studies of immune dysregulation and potential therapeutic interventions in equine health.

The kit is compatible with serum, plasma, cell culture supernatant, cell lysis buffer or tissue lysis buffer, and other biological fluid samples. Reported performance characteristics include a sensitivity of 9.375 pg/mL, an assay range of 15.625-1000 pg/mL, and a sandwich ELISA, double antibody format with colorimetric readout. Linearity across serial dilutions (1:2, 1:4, 1:8) ranges from 86-101% for serum, 88-102% for EDTA plasma, and 91-100% for heparin plasma (all n=5), with average recovery values of 93% for serum, 95% for EDTA plasma, and 95% for heparin plasma. The kit should be stored at 2-8°C and remains stable for 12 months under these conditions.

The assay workflow follows a standard sandwich ELISA procedure: equilibrate reagents and TMB substrate to room temperature, then set up standards, samples and controls on the pre-coated plate; incubate at 37°C for 90 minutes for antigen binding; add biotin-labelled detection antibody and incubate at 37°C for 60 minutes; add HRP-Streptavidin (SABC) and incubate at 37°C for 30 minutes; develop with TMB substrate in the dark for 10-20 minutes; stop the reaction and read absorbance at 450 nm immediately.
