Description

Assay Genie provides a Cell-Based ELISA Kit designed for the qualitative detection of hnRNP K phosphorylation at serine 284 in cell lysates and tissue homogenates. The kit is configured for adherent cell samples, employs an indirect ELISA format with colorimetric readout at 450 nm, has a dynamic range of > 5000 Cells, and is reactive across human, mouse and rat samples. It is positioned for immunology research to measure phosphorylated hnRNP K levels and to study the effects of various treatments, inhibitors or activators on hnRNP K phosphorylation in cultured cells.

The assay is lysate-free and high-throughput, using target-specific primary antibodies paired with HRP-conjugated secondary antibodies that catalyze a colorimetric reaction upon substrate addition. A monoclonal antibody specific for human GAPDH is included to serve as an internal positive control for normalizing the target absorbance values. A Crystal Violet whole-cell staining method is also provided to determine cell density, allowing absorbance values to be normalized to cell amounts so plating differences can be adjusted.

hnRNP K is a heterogeneous nuclear ribonucleoprotein involved in RNA processing and gene expression regulation. It binds tenaciously to poly(C) sequences and is described as playing a role in the nuclear metabolism of pre-mRNAs containing cytidine-rich sequences. Phosphorylation at serine 284 is described as playing a critical role in various cellular processes, making the protein a valuable biomarker for studying signaling pathways and disease mechanisms relevant to cancer and neurodegenerative disorders. The associated gene is HNRNPK (Gene ID: 3190, UniProt ID: P61978, OMIM: 600712), located at chromosomal position 9q21.32, with a molecular weight of 48,562 Da.
