Description

From Assay Genie, this cell-based ELISA kit is designed for the qualitative detection of HNF4alpha phosphorylated at serine 313, supporting measurement of phosphorylation status and total protein expression in cultured cells. The assay uses a lysate-free, high-throughput indirect ELISA format in which HNF4alpha (Phospho-Ser313)-specific primary antibodies are bound by HRP-conjugated secondary antibodies, with HRP activity read colorimetrically at 450 nm after substrate addition.

Reactivity is reported for human, mouse and rat samples, and the workflow is intended for adherent cell cultures. A dynamic range of > 5000 Cells is stated. A monoclonal anti-GAPDH antibody is included as an internal positive control for normalising target absorbance, and Crystal Violet whole-cell staining can be used to assess cell density and correct for plating differences.

HNF4alpha (gene symbol HNF4A) is a nuclear transcription factor that binds DNA as a homodimer and regulates hepatic genes including hepatocyte nuclear factor 1 alpha, apolipoprotein CIII and transthyretin. The kit is suited to cell biology, molecular biology and drug development research, where phosphorylation at Ser313 is of interest in relation to liver development, glucose and lipid metabolism, and diseases such as diabetes, liver dysfunction and cancer.
