HNF4alpha Colorimetric Cell-Based ELISA Kit

From Assay Genie, this cell-based ELISA kit enables direct measurement of HNF4alpha protein expression in cultured cells without a lysate preparation step. The assay uses an indirect ELISA format in which HNF4alpha is captured by target-specific primary antibodies, and bound primary antibodies are detected using HRP-conjugated secondary antibodies that catalyze a colorimetric reaction read at 450 nm. The kit is reactive with Human, Mouse, and Rat samples, is intended for use with adherent cells, and has a stated dynamic range of > 5000 Cells. A monoclonal antibody specific for human GAPDH is included to serve as an internal positive control, and a Crystal Violet whole-cell staining step is provided for normalizing absorbance values to cell density. HNF4alpha (gene symbol HNF4A) is a nuclear transcription factor that binds DNA as a homodimer. It controls the expression of several hepatic genes, including hepatocyte nuclear factor 1 alpha, alpha 1-antitrypsin, apolipoprotein CIII, and transthyretin, and has been described as potentially essential for development of the liver, kidney, and intestine. Mutations in the gene have been associated with monogenic autosomal dominant non-insulin-dependent diabetes mellitus type I. The kit is suited to measuring relative HNF4alpha levels in cultured cells and to screening the effects of various treatments, inhibitors (such as siRNA or chemicals), or activators on HNF4alpha expression.
