Description

From Assay Genie, this Cell-Based ELISA kit enables sensitive and accurate detection of HKR1 protein levels in cell lysates and culture supernatants, with applications in research on cell migration, invasion, metastasis, and tumor progression. Relevant to the Epigenetics and Nuclear Signaling research area, the kit provides high specificity and reproducibility for human samples using adherent cells.

The assay uses a 96-Well Microplate format with colorimetric detection at 450 nm and a dynamic range of > 5000 Cells. Storage is at 4°C with a stated stability of 6 Months. As a cell-based, lysate-free, high throughput and sensitive assay, the kit can detect the HKR1 protein expression profile in cultured cells and is used to measure the relative amounts of HKR1, as well as to screen the effects of treatments, inhibitors such as siRNA or chemicals, or activators on HKR1.

Qualitative determination of HKR1 concentration is achieved by an indirect ELISA format: HKR1 is captured by HKR1-specific primary antibodies, and HRP-conjugated secondary antibodies bind the Fc region of the primary antibody. The HRP enzyme then catalyzes a colorimetric reaction upon substrate addition. A monoclonal antibody specific for human GAPDH is included as an internal positive control for normalizing target absorbance values, and a Crystal Violet whole-cell staining step may be used to determine cell density and adjust for plating differences.
