Histone H1 (Phospho-Thr17)Fluorometric Cell-Based ELISA Kit

From Assay Genie, this Fluorometric Cell-Based ELISA Kit is designed for the qualitative determination of phosphorylated Histone H1 at threonine 17 in cultured cells. It is described as a convenient, lysate-free, high-throughput and sensitive assay that can monitor Histone H1 phosphorylation and expression profile in cells. The kit can be used for measuring the relative amounts of phosphorylated Histone H1 (Phospho-Thr17) in cultured cells, as well as for screening the effects that various treatments, inhibitors (such as siRNA or chemicals), or activators have on Histone H1 phosphorylation.

Detection is achieved by an indirect ELISA format. Phospho-Histone H1 (Thr17) is captured by target-specific primary antibodies, while dye-conjugated secondary antibodies bind the Fc region of the primary antibody. The bound dye emits light at defined wavelengths upon proper excitation, enabling fluorometric readout at Ex/Em: 651/667 (Dye 1) and 495/521 (Dye 2). The kit is reactive with Human and Mouse samples, with a stated dynamic range of > 5000 Cells.

A monoclonal antibody specific for human GAPDH is included to act as an internal positive control in normalizing the target RFU values, and an antibody against the non-phosphorylated counterpart of Histone H1 (Phospho-Thr17) is also provided for normalization purposes.
