Description

A cell-based ELISA kit from Assay Genie for the qualitative detection of HINT1 (Histidine triad nucleotide-binding protein 1) protein expression in cultured cells. The assay uses an indirect ELISA format with HINT1-specific primary antibodies and HRP-conjugated secondary antibodies, providing a lysate-free, high-throughput workflow suited to adherent cell samples. Detection is colorimetric, read at 450 nm, with absorbance results normalised using an included anti-GAPDH monoclonal antibody as an internal positive control and Crystal Violet whole-cell staining to correct for plating differences.

The kit is reactive with Human, Mouse and Rat samples and supports a dynamic range of > 5000 Cells, making it suitable for measuring relative HINT1 levels and for screening the effects of treatments, inhibitors such as siRNA or chemicals, and activators. It is supplied in a microplate format with a storage recommendation of 4°C for 6 Months, and is positioned within the Cell Death research area.

HINT1 is a hydrolase of the HINT family that hydrolyzes adenosine 5'-monophosphoramidate substrates such as AMP-morpholidate, AMP-N-alanine methyl ester, AMP-alpha-acetyl lysine methyl ester and AMP-NH2. The encoded protein has been identified as a tumor suppressor gene and is associated with Neuromyotonia and Axonal Neuropathy, Autosomal Recessive. It participates in transcription regulation, signal transduction and purine ribonucleotide catabolism, and localises to the cytoplasm, nucleus, cytoskeleton, histone deacetylase complex and plasma membrane.
